The determination of high-resolution structural models of macromolecules, obligatory for detailed structure–function studies, requires large amounts of pure, stable target protein. The two principal methods of structure determination are X-ray crystallography, a method that requires growing highly ordered 3D crystals of the target protein, and nuclear magnetic resonance (NMR) spectroscopy, a method that requires the generation of doubly isotopically labeled (15N and 13C) protein.
